چکیده

The present work describes the optimization of a short-term assay, based on the inhibition of the esterase activity of the alga Pseudokirchneriella subcapitata, in a microplate format. The optimization of the staining procedure showed that the incubation of the algal cells with 20 μmolL fluorescein diacetate (FDA) for 40 min allowed discrimination between metabolic active and inactive cells. The shortterm assay was tested using Cu as toxicant. For this purpose, algal cells, in the exponential or stationary phase of growth, were exposed to the heavy metal in growing conditions. After 3 or 6 h, cells were subsequently stained with FDA, using the optimized procedure. For Cu, the 3and 6-h EC50 values, based on the inhibition of the esterase activity of algal cells in the exponential phase of growth, were 209 and 130 μg L, respectively. P. subcapitata cells, in the stationary phase of growth, displayed higher effective concentration values than those observed in the exponential phase. The 3and 6-h EC50 values for Cu, for cells in the stationary phase, were 443 and 268 μgL, respectively. This short-term microplate assay showed to be a rapid endpoint for testing toxicity using the alga P. subcapitata. The small volume required, the simplicity of the assay (no washing steps), and the automatic reading of the fluorescence make the assay particularly well suited for the evaluation of the toxicity of a high number of environmental samples.

Responses of the alga Pseudokirchneriella subcapitata to long-term exposure to metal stress.

The green alga Pseudokirchneriella subcapitata has been widely used in ecological risk assessment, usually based on the impact of the toxicants in the alga growth. However, the physiological causes that lead algal growth inhibition are not completely understood. This work aimed to evaluate the biochemical and structural modifications in P. subcapitata after exposure, for 72 h, to three nominal ...

Toxicity assessment of pesticides to Pseudokirchneriella subcapitata under air-tight test environment.

This paper presents the toxicity data of seven pesticides including atrazine, parathion, dichlorvos, malathion, fenthion, 2-methyl-4-chlorophenoxyacetic acid, and pentachlorophenol on Pseudokirchneriella subcapitata based on a new algal toxicity testing technique conducted under air-tight environment. The dissolved oxygen production and the cell density were adopted as the response endpoints. M...

Assessment of catalase activity, lipid peroxidation, chlorophyll-a, and growth rate in the freshwater green algae Pseudokirchneriella subcapitata exposed to copper and zinc

In this work, the effect of copper and zinc on green alga Pseudokirchneriella subcapitata was evaluated through catalase activity, lipid peroxidation by TBARS essay, growth rate, and the chlorophyll-a concentration. Catalase activity increased significantly (P < 0.05) in comparison to the control at 0.1 mg L copper and 0.075 mg L zinc, whereas the damage to the cell membrane expressed as nmols/...

A Sensitive Neutralization Assay for Influenza C Viruses Based on the Acetylesterase Activity HEF Glycoprotein

Influenza C virus possesses specific neuraminate-O-acetylesterase as a receptor-destroying function. This enzymatic activity of the viral glycoprotein HEF (Hemagglutinin, esterase activity and fusion factor) can be visualized in situ by the use of distinct color substrates. Hereby the localization, as well as the quantity of synthesized HEF protein is detectable. We further developed the estera...

Toxicity of chlorophenols to Pseudokirchneriella subcapitata under air-tight test environment.

A closed-system algal toxicity test with no headspace was applied to evaluate the toxicity of chlorophenols to Pseudokirchneriella subcapitata. The dissolved oxygen production and the growth rate based on cell density were the response endpoints. Phenol and seven chlorophenols were tested using the above test technique. Median effective concentrations (EC50) range from 0.004 to 25.93 mg/l (base...
